Anti-CD19 CAR-T with IL-6 Knockdown for CNS B-ALL
Phase 1 clinical trial (NCT03064269) of ssCART-19s targeting CD19 with shRNA-IL-6 gene silencing for CNS B-cell ALL. Three patients achieved near-complete symptom resolution, significant brain infiltration reduction on MRI, and negative CSF blasts. Only grade 1 CRS observed. PubMed: 32745961

Analysis of 56 adults with r/r Ph+ ALL: 91.1% CR/CRi with humanized CD19 CAR-T. Consolidative alloHSCT improved 2-year OS (58.9% vs 22.7%) and LFS (53.2% vs 18.8%). MRD-negative post-CART was independent prognostic factor. PMC: 13063648
